Hephaestin is a protein encoded by the HEPH gene in humans. It is referred as HEPH and CPL. It is a multicopper ferroxidase involved in iron absorption in the small intestine. The ferroxidase activity of this protein may play an important role during iron export from intestinal enterocytes and the subsequent iron loading of the blood protein transferrin delivers iron to the tissues. It is expressed mainly on the basolateral surface of duodenal enterocytes, facilitating the export of iron from the intestinal epithelium into blood by oxidizing Fe²⁺ into Fe³⁺, the only form of iron bound by the plasma protein transferrin.
