The gene HINT2 (histidine triad nucleotide-binding protein 2, mitochondrial) is mapped to human chromosome 9p13.2. It belongs to the histidine triad hydrolase enzymes superfamily. HINT2 is strongly expressed in liver and pancreas. The protein localizes in the mitochondria and contains a conserved HIT motif (histidine-X-histidine-X-histidine).

HINTs (histidine triad nucleotide-binding proteins) are mainly adenosine monophosphate-lysine hydrolases. HINT2 regulates the mitochondrial-dependent apoptosis. It is down-regulated in hepatocellular carcinoma. HINT2 is suggested as a tumor suppressor for endometrial cancer. It also participates in steroidogenesis.
