Ring-box 1 (RBX1) gene encodes an evolutionarily conserved protein that interacts with cullins. The protein plays a unique role in the ubiquitination reaction by heterodimerizing with cullin-1 to catalyze ubiquitin polymerization. It also may be involved in the regulation of protein turn-over (provided by RefSeq). Ring-box 1 (RBX1) gene is located on human chromosome 22q13.2. RBX1 is a component of SCF (Skp1, Cullins, F-box proteins) E3 ubiquitin ligases. RBX1 is a14 kDa protein with a ring finger domain (C₃H₂C₃), essential for ubiquitin ligation.

Ring-box 1 (RBX1) mediates neddylation of cullin-1(CUL1). RBX1 regulates protein turnover. RBX1 is a potential anticancer target. Mutations in RBX1 is linked to tumorigenesis and tumor progression. RBX1 acts as a drug target for esophageal cancer. RBX1 has a role in bladder urothelial cell carcinoma (UCC) carcinogenesis and progression.
