| Biochem/physiol Actions【生化/生理作用】 | iP300, a mixture of 8 diastereomers, is a potent histone acetyltransferase (HAT) p300/CBP inhibitor (p300 IC50 = 33 nM by H3K9Ac HTRF assay). An active diastereomer(s)-enriched pool (iP300w) obtained by partial HPLC separation prevents DUX4-induced cytotoxicity and transcription activity in human myoblast LHCN-M2 cultures (0.1-1 μM) and shows in vivo efficacy against muscle loss (0.3 mg/kg/day i.p.) in a murine model of facioscapulohumeral muscular dystrophy (FSHD). |
